Arbuscular mycorrhizal-like fungi (AM-like fungi) are crucial for ecosystem functioning and soil organic matter (SOM) is an indicator of soil quality. However, the spatial distribution of arbuscular mycorrhizal-like fungi, glomalin-related soil protein (GRSP) and SOM in a large scale is still unclear. The objectives of this study were to investigate the spatial distribution of SOM, arbuscular mycorrhizal-like fungi and GRSP, and reveal the potential relationship among them in a large scale across China.
Materials and methods
Soil samples (different in vegetation type, climate, and soil variables) were collected from 26 sites in a large scale across China. The soil properties including pH, total carbon (TC), total nitrogen (TN), and SOM were determined. Quantitative PCR amplification of the 18S rRNA gene was conducted to evaluate the abundance of arbuscular mycorrhizal-like fungi. The contents of easily extractable GRSP (EE-GRSP), difficultly extractable GRSP (DE-GRSP), and total GRSP (T-GRSP) were measured.
Results and discussion
Arbuscular mycorrhizal-like fungi abundance was significantly affected by the vegetation type and dramatically correlated with the soil TN and mean annual precipitation (MAP). EE-GRSP and DE-GRSP were more associated with the TC and TN content, respectively. The abundance of arbuscular mycorrhizal-like fungi significantly but weakly correlated with the T-GRSP and EE-GRSP. The SOM content positively correlated with the DE-GRSP and T-GRSP. Those results suggested that the arbuscular mycorrhizal-like fungi are a larger contributor to regulating the content of GRSP, which is an important indicator of the soil organic carbon pool.
Conclusions
Our results indicated that arbuscular mycorrhizal-like fungi abundance has a greater contribution to driving the distribution of soil C and N in a large scale by affecting the content of glomalin-related soil protein.
l-proline (Pro) is a precursor of ornithine, which is converted into polyamines via ornithine decarboxylase (ODC). Polyamines plays a key role in the proliferation of intestinal epithelial cells. The study investigated the effect of Pro on polyamine metabolism and cell proliferation on porcine enterocytes in vivo and in vitro. Twenty-four Huanjiang mini-pigs were randomly assigned into 1 of 3 groups and fed a basal diet that contained 0.77% alanine (Ala, iso-nitrogenous control), 1% Pro or 1% Pro + 0.0167% α-difluoromethylornithine (DFMO) from d 15 to 70 of gestation. The fetal body weight and number of fetuses per litter were determined, and the small and large intestines were obtained on d 70 ± 1.78 of gestation. The in vitro study was performed in intestinal porcine epithelial (IPEC-J2) cells cultured in Dulbecco''s modified Eagle medium-high glucose (DMEM-H) containing 0 μmol/L Pro, 400 μmol/L Pro, or 400 μmol/L Pro + 10 mmol/L DFMO for 4 d. The results showed that maternal dietary supplementation with 1% Pro increased fetal weight; the protein and DNA concentrations of the fetal small intestine; and mRNA levels for potassium voltage-gated channel, shaker-related subfamily, member 1 (Kv1.1) in the fetal small and large intestines (P < 0.05). Supplementing Pro to either gilts or IPEC-J2 cells increased ODC protein abundances and polyamine concentrations in the fetal intestines and IPEC-J2 cells (P < 0.05). In comparison with the Pro group, the combined administration of Pro and DFMO reduced the expression of ODC protein and spermine concentration in the fetal intestine, as well as the concentrations of putrescine, spermidine and spermine in IPEC-J2 cells (P < 0.05). Meanwhile, the percentage of cells in the S-phase and the mRNA levels of proto-oncogenes c-fos and c-myc were increased in response to Pro supplementation, whereas depletion of cellular polyamines with DFMO increased tumor protein p53 (p53) mRNA levels (P < 0.05). Taken together, dietary supplementation with Pro improved fetal pig growth and intestinal epithelial cell proliferation via enhancing polyamine synthesis. 相似文献
This study was designed to examine the effects of glycinin on growth, digestive ability, immune responses, antioxidant capacity and gene expression levels of golden crucian carp. Golden crucian carp were fed diets containing glycinin at 0, 30, 60, 90 and 120 g/kg, respectively, for 8 weeks. Body weight, weight gain percentage, specific growth rate and feed efficiency ratio were negatively related to the content of glycinin in diet. Activities of protease, acid phosphatase, alkaline phosphatase, lysozyme in hepatopancreas, and activities of catalase, glutathione peroxidase, superoxide dismutase, and total antioxidant capacity in the proximal intestine, mid intestine, distal intestine and hepatopancreas were negatively related to the content of glycinin in diet, whereas malondialdehyde in proximal intestine, mid intestine, distal intestine and hepatopancreas increased directly with the content of glycinin in diet. Furthermore, the relative expressions of TNF‐α and IL‐1β in proximal intestine, mid intestine and distal intestine increased directly with the content of glycinin in diet, whereas the relative expressions of TNF‐α and IL‐1β in hepatopancreas were negatively related to the content of glycinin in diets. The relative expressions of IL‐10 in proximal intestine, mid intestine, distal intestine and hepatopancreas all were negatively related to the content of glycinin in diets. In conclusion, reductions in growth, immunity and antioxidant capacity, intestine inflammation with dysfunction of digestive system occurred in golden crucian carp that fed a diet containing glycinin at 30 g/kg or higher after 8 weeks. 相似文献
